PURINE ALKYLATING AGENTS 2 , 6-DIAMINO-8-HALO ALKYL PURINES (POTENTIAL ANTI - MALARIALS)

Authors: not saved
Abstract:

Haloalkyl purines i.e. 2,6-diamino-8-(chloromethyl) purine 10; 2, 6-diamino-X- (chloromethyl) purine 17; 2,6-diamino-8-(chloropropylp) urine 18; were synthesized by two distinct but facile methods. The N-alkylating potential of 2, 6-diamino-8- (chloromethyl) purine was investigated. The following products 2,6-diamino-8-(Nbenzyl- N-methyl) purine 11; 2,6-diamino-8-(4-nitro anilinomethyl) purine 13; 2,6- diarnino-8-(piperidino-N-methyl)Purine 14; 2,6-diamino-8-(3,4,5-trimethoxyanilinomethyl) purine 12; of N-alkylation are reported. Antifolate activities obtained for these compounds are also reported. 2,6-diamino-8-(3,4,5-trimethoxyanilinom ethyl) purine demonstrated significant biological activity (ID = 1 x10 ). Most antifols are also antimalarials e.g. trimethoprim and pyrimethamine, and therefore these 8-haloalkyl purines may be effective antimalarials if modified by appropriate reagents

Purines, Purine Nucleosides, and Analogues

Of 142 purines, purine nucleosides, and analogues tested for inhibition of growth of Escherichia coli B Hill, 45 were active. Of these, 27 were evaluated for inhibition of other E. coli lines, including those resistant to 6-thioguanine, 2-fluoroadenosine, 2,6-diaminopurine, or 6-mercaptopurine. Most toxic to the parent lines were 2-fluoroadenosine, 2-fluoroadenine, 2-fluoro-5'-deoxyadenosine, a...

6-(2-Methoxybenzylamino)purine

The title compound, C(13)H(13)N(5)O, consists of discrete mol-ecules connected by N-H⋯N hydrogen bonds to form infinite chains, with N⋯N separations of 3.0379 (15) and 2.8853 (15) Å. The benzene and purine ring systems make a dihedral angle of 77.58 (3)°. The crystal structure is further stabilized by intra-molecular N⋯O inter-actions [2.9541 (12) Å] and inter-molecular C-H⋯C and C⋯C contacts [...

Microwave promoted C6-alkylation of purines through S(N)Ar-based reaction of 6-chloropurines with 3-alkyl-acetylacetone.

C6-Alkylated purine analogues were obtained in good to excellent isolated yields by S(N)Ar reaction of 6-chloropurine derivatives with 3-alkyl-acetylacetone. 3-Alkyl-acetylacetones were employed as alkylating agents and C6-alkylated purines were obtained highly selectively within short reaction time under microwave irradiation conditions. This work is complementary to the classical coupling rea...

The Synthesis of 2, 6 diamino- 3, 5-dinitropyridine from Direct Nitration Reaction of 2, 6 diamino pyridine in Different Temperature Conditions by Density Functional Theory

In this article, synthesis of the explosive 2, 6 (diamino) 3, 5 dinitro pyridine (ANPY) of the direct nitration reaction of 2, 6 diamino pyridine in different conditions of temperature, with density functional theory method were studied. For this purpose, at first the material contained in the both sides of reaction were geometrically optimized, then the calculation of the the...

Novel Oxadiazole Thioglycosides as Potential Anti- Acinetobacter Agents

The glycosylation of 1,3,4-oxadiazole-2-thiones has been performed with peracetylated β-pyranosyl bromide in the presence of potassium carbonate. Deprotection of acetylated thioglycosides was necessary for increasing their antibacterial effects. The structure of nucleosides were confirmed by 1H NMR, 13C NMR and HRMS. The anomeric protons of nucleosides c1–4 were assigned to the doublet, confirm...
